Principle of the separation procedure
Prior to Lymphoprep, the most common technique for separating white blood cells is to mix blood with a compound that aggregates red blood cells, thereby increasing their sedimentation rate. Leukocyte sedimentation is only slightly affected and can be collected from the top of the tube when the RBCs have settled.
Using a mixture of sodium metrizoate and polysaccharide, the initial version of Lymphoprep™, Bøyum (1968) developed a one-step centrifugal technique for the isolation of lymphocytes. Thorsby and Bratlie (1970) used this technique with slight modifications in the preparation of a suspension of pure lymphocytes for cytotoxicity tests and lymphocyte cultures. As also emphasized by other authors, Harris and Ukayiofo (1969), and Ting and Morris (1971), Lymphoprep™ is a reliable, simple and rapid method suitable for the preparation of lymphocyte preparations from cadaver blood and stored anticoagulated blood. at room temperature. up to 6 hours.
